Test Strip Instructions
Ⅰ. Composition:
1. The QTY of MCMV test strip: 50 pcs
2. Plant tissue grinding bag: 50 pcs
3. Sample extraction buffer: 50mL 1 bottle
Ⅱ. Operating Steps:
1. Sample preparation:
1.1 Method 1 of Plant Sample Preparation:
(1) Collection of plant samples: Take a leaf (30-50mg) about half the size of a thumb nail from a watermelon leaf with hand or scissors and put it into the grinding plastic bag provided.
(2) Grinding and homogenizing of plant samples: Add 2 tubes of sample extraction solution (about 1 ml) to the grinding bag containing leaves with the provided straw, rub the plastic bag with both hands like washing clothes until the leaves are completely crushed, and mix in the sample extraction solution.
1.2 Method 2 of Plant Sample Preparation:
(1) Collect Corn leaves with obvious seedling or Mosaic symptoms, and weigh about 30-50 mg of leaf tissue into a mortar.
(2) Add 1 ml of the extract into the mortar and grind the homogenate with a pestle for 1 minute.
(3) The homogenate is transferred to a 1.5ml plastic centrifuge tube, centrifuged at 5000 RPM for 3 minutes, or left for 3 minutes, and the supernatant is the sample detection liquid.
1.3 Seed Sample Preparation:
(1) Separation of seed kernel and seed shell of seed sample: Cut a corn seed along the side with scissors, peel off the seed kernel part, collect the remaining seed shell and put it in a mortar.
(2) Grinding and homogenizing of seed shells: Suck 1 tube of sample extract liquid with a straw and add it to a mortar with 1 seed shell, and grind the homogenization with a pestle for 5 minutes.
2.Sample Test:
Take out the test strip from the aluminum foil bag, lay it flat face up as shown in the figure below, and use a straw to absorb the above prepared sample extraction drops 3-4 drops into the sample adding hole.

3. Result Interpretation:
The results were observed within 3 to 10 minutes. As shown in Figure 1 below, and the color was positive at both line C (control line) and line T (detection line). As shown in Figure 2 below, color rendering only on line C and no color rendering on line T is negative. If neither line C nor line T is colored, or only line T is colored and line C is not colored, the test strip is invalid.

Ⅲ. Notes:
1. When testing plants, the amount of diseased leaves should not be much, the sampling amount should be strictly controlled, half or one thumbnail size is enough, if too many leaves are taken, the green leaf in the leaves will affect the detection color, and the detection sensitivity of the test strip is very high, there will be no false negative phenomenon caused by insufficient sample amount.
2. Store the test strip in a dry sealed aluminum foil bag and use it now.
3. Test strips are used for MCMV detection.
4. Watermelon leaves with obvious virus symptoms should be collected during sampling. The symptoms of MCMV infection in melon plants include: irregular fading of young leaves or light yellow flowers and leaves, followed by bumps in the green part of the leaf surface, but the symptoms gradually reduce as the leaves age.
5. Take samples by hand, scissors or blade, and clean them with running water after each sample is taken.
6. The colloidal gold test strip must be used with the sample extraction solution provided, and other sample extraction solutions cannot be used.
7. Each straw can only be used for the absorption and addition of 1 sample, and it is prohibited to use the same straw for the absorption and addition of multiple samples.
Ⅳ. Storage conditions:
Store test strips in a cool dry environment away from light at 4-30℃. Do not freeze.
Ⅴ. Production Date and Expiration Date:
Production dates are detailed in the box. It is valid for 12 months.
