Colloidal Gold Immunoassay Strip Instructions of Acidovorax Citrulli (Aac)

Ⅰ. Testing Principle:

The colloidal gold immune test strip for the detection of acidovorax citrulli is a qualitative test strip. The paper strip is based on the formation of double antibody sandwich immune complex on the detection line of nitrocellulosic membrane to detect the specific and sensitive acidovorax citrulli.

 

Ⅱ. Range of Application

This product is used for rapid detection of acidovorax citrulli in leaf or seed samples of cucurbitaceae plants.

Ⅳ. Operating Steps:

(1)    Sample Preparation:

Melon leaf sample: Add all the extract (1 mL in total) from the sample tube into the grinding bag, then add 0.1g leaves with obvious symptoms into it, rub the grinding bag with your hands like rubbing clothes until the leaves are completely crushed, and the homogenate obtained is the sample detection solution.

Seed samples: Randomly take 5 seeds from each sample and put them into a sample processing tube containing 1 mL of extraction solution, and shake them at room temperature for 20-30 min; Or cut 5 seeds along the side with scissors, peel off the kernel part, collect the remaining seed shells in a sample processing tube containing 1 mL of extraction solution, and shake at room temperature for 20-30 min (recommended).

(2)    Sample Test:

Remove the strip from the aluminum foil bag and lay it flat face up. Use a plastic straw to absorb the leaf homogenate or seed extract. Add 3-4 drops (about 100µL) into the sampling hole (S) of the strip and place it for 5-10 minutes to judge the result.

Negative (-) : Line C shows color, line T does not show color, and is judged as negative.


Positive (+) : C line color, T line also color, regardless of the color of the T line is judged to be positive.

Invalid: No C line appears, regardless of whether the T line is colored or not, indicating an incorrect operation process or the detection strip has failed. In this case, the instructions should be read carefully again and retested with a new test strip.

 

Ⅵ. Notes:

(1) Perform the test according to the procedure. Do not touch the color development area of the test strip during operation.

(2) This product is a disposable product, do not reuse.

(3) Do not use tap water, pure water, distilled water and normal saline as a negative control or sample extraction solution.

(4) Do not mix plastic straws from different samples.

(5) After taking out the test strip from the original packaging aluminum foil bag, it should be used within 1 hour.

(6) Place the test strip on a flat table for testing, and add 3-4 drops of sample extraction solution vertically.

(7) Because the bacteria will precipitate at rest, the sample extract is slightly mixed before the plastic straw is sucked.

(8) Due to differences between samples, T, C line color glossiness will be different, which is a normal phenomenon.

(9) Too low ambient temperature in the experiment will lead to abnormal test strips, please keep the ambient temperature at 4-37℃.

 

VII. Storage Condition and Validity:

Storage conditions: Store test strips in a cool dry environment away from light at 4-30℃. Do not freeze.

Validity: The product is valid for 24 months. The date of manufacture is on the box.
